Titmus' second place in Saturday's final won by American Katie Ledecky gives the Australian a fourth medal of the Games.
It follows her wins in the 200m and 400m freestyles and a bronze in the 4x200m freestyle relay.
In the 800m freestyle final, Titmus and the rest of the field were unable to counter Ledecky, who won the event for a third consecutive Olympics to claim a seventh career gold medal.
"I knew this would be the toughest race for me," Titmus said.
"I definitely think I'm better a 200 and 400 swimmer than an 800 swimmer ... I knew it would be tough so I'm happy with the silver medal.
"She (Ledecky) is pretty much in a class of her own in the 800."
The American won in eight minutes 12.57 seconds from Titmus (8:13.83) and Italian Simona Quadarella (8:18.35).
Australia's Kiah Melverton (8:22.25) finished sixth.
Titmus will now take a break after a wildly successful debut Olympics.
"Relieved and kind of sad that my Tokyo campaign is over but I'm happy how I performed this week," she said.
"I would like to have a good break now ... I need time to let my mind and body recover and my shoulder needs a good break.
"I really do have to think about what I've achieved and have time away from the pool so I can get ready and up for next year."
