Former vice-captains Josh Kelly and Stephen Coniglio have made way for Green, alongside defenders Connor Idun and Jack Buckley in the four-man leadership group.
Veteran midfielder Kelly, 30, relinquishes his vice-captaincy as he looks to make his return from hip surgery mid-way through the season.
One of two inaugural Giants left alongside Greene, the 32-year-old Coniglio is out to make up for lost time on the field after a torrid injury run limited him to just 25 games across the past two seasons.
Greene enters his fourth season as captain under coach Adam Kingsley, after sharing the role with Kelly and former stand-alone skipper Coniglio.
GWS football boss Jason McCartney said it made "perfect sense" for 2023 All-Australian captain Greene to continue as the sole skipper, while praising Kelly and Coniglio.
"Over the last three years, he's (Greene) led our side brilliantly, and we have no doubt he'll continue to do the same this year," McCartney said in a club statement.
"I'd also like to acknowledge the tremendous work and leadership of our two outgoing vice-captains, Josh Kelly and Stephen Coniglio.
"The work they've done as leaders of our club is immeasurable, and while they don't have the title anymore, we know they'll both continue to be incredible leaders.
"Both Stephen and Josh are very supportive of the change and the opportunity that it presents for Tom, Connor and Jack."
Green has long been touted as a future captain of GWS, and has been part of the club's leadership alongside Idun for the past three years.
Buckley was elevated to the leadership group for the first time last year, with injured small forward Brent Daniels and star defender Sam Taylor dropping out of the set-up under Kingsley.
"It's also great to have Tom, Connor and Jack take on expanded leadership roles and be elevated to vice-captains," McCartney said.
"All three continue to show exceptional leadership qualities and it's great to have them continue to develop as leaders under Toby's guidance.
"Our leadership for 2026 brings together a strong mix of experience and fresh energy, and we're excited about where it can take the club in 2026 and beyond."
